Communicating the Future: Engaging the Public in Basic Science will explore why the public and basic science should be connected and what current communication and engagement efforts are taking place\, including the challenges and opportunities in this work. Ultimately\, we intend to identify how scientists and professionals can work to engage the public with basic research as effectively as possible. \nThe conference program will be organized around the exploration of the why\, what and how of the relationship between the public and basic science. The following aspects will be explored throughout: \nScholarship (e.g. communications research) – whether and how it can be applied to public engagement practice\, and identification of new research questions; \nTraining – public engagement skills development for scientist-communicators and the communication professionals who work with them; \nEngagement practice – activities or programs that provide opportunities for mutual learning or exchange of ideas between scientists and members of the public; \nJustice\, equity\, diversity\, and inclusion in public engagement; and \nEvaluating the effectiveness of public engagement activities and programs. \nWhat we discuss and learn at this conference will also inform future directions of SciPEP and the resources and training we propose to develop for basic scientists. \nPlenary \nHow do we move forward from here? \nAn astrophysicist\, a political scientist\, and science journalist turned college dean walk into a public engagement conference\, and… what comes next? Myelin development and homeostasis are critical for normal function and impairment of myelin is associated with sensory\, motor\, and cognitive dysfunction. While it has long been understood that oligodendrocytes produce myelin in the CNS\, more recent studies also implicate additional cell types including microglia as providers of important molecular cues in myelination. However\, the function of many molecular mediators remains incompletely defined\, at least in part due to the scarcity of suitable tools. In this thesis\, we develop and provide the research community with much needed new tools to study microglia and myelin and dissect the contribution of IgG to myelin development. In Chapter 1\, we report the generation and characterization of now widely adopted EGFP and CreERT2 transgenic mouse lines for the labeling and the inducible genetic manipulation of microglia. Extending this work\, in Chapter 2\, we present evidence for the engineering of a highly efficient and specific constitutively active Cre line leveraging the Fcrls locus. In Chapter 3\, we report the development of MyelTracer\, an easy-to-install software suite made available to the research community for the quantification of myelin g-ratio\, a key metric in studies of myelin morphology. Finally\, in Chapter 4\, we present evidence for the occurrence of maternally derived IgG on microglia in the postnatal brain and show that brains of mice lacking IgG postnatally display oligodendroglial and axonal abnormalities. Further\, we show that this effect of IgG is not mediated through the canonical IgG-Fc receptor pathway. Independent of Fc receptors\, IgG appears to be required for the function of a subset of microglia that occurs in white-matter tracts postnatally and is required for normal white-matter development. Overall\, the work presented in this thesis resulted in the generation of several much-needed tools for the research community\, and it reveals molecular insights into the role of IgG in the developing brain.

SUMMARY:Dana-Farber Targeted Protein Degradation Webinar Series: Dr. Stuart Schreiber
DESCRIPTION:30 years of Molecular Glues: Controlling cell circuitry in biology and medicine\n\nHost: Alyssa Verano \n\n\nDr. Schreiber’s research integrates chemical biology and human biology to advance both our understanding of chemistry and biology\, and the discovery of novel therapeutics. He is known for his use of small molecules to explore biology and medicine\, and for his role in the development of the field of chemical biology. With Jerry Crabtree in 1991\, his lab mapped the first membrane to nucleus signaling pathway (calcium–calcineurin–NFAT). His lab co-discovered mTOR in 1994 (simultaneously with Sabatini) and helped illuminate the mTOR-dependent nutrient-response signaling network. His lab discovered histone deacetylase (HDAC) and\, together with David Allis and Michael Grunstein in 1996\, the role of chromatin marks in gene expression. His work demonstrated for the first time that drugs can result from: 1) the targeting of protein kinases (sirolimus/mTOR) and protein phosphatases (sandimmune/ calcineurin); 2) gene regulation by chromatin-modifying enzymes (vorinostat/HDAC)\, 3) chemical inducers of proximity (CIPs) that activate cellular processes by enforced proximity (GVH Disease)\, and 4) targeting of the proteasome (bortezomib/proteasome). His research illustrated that many small molecules are bifunctional and act by inducing proximity of signaling proteins\, which he coined ‘molecular glues’. The discovery of molecular glues and development of chemical inducers of proximity led conceptually to the targeted degradation of proteins by small-molecule ‘PROTACs’. These efforts accelerated the development of many additional widely used drugs and more generally the field of chemical biology. \nSchreiber’s development of diversity-oriented synthesis has led to the discovery of many promising agents\, including a novel mechanism of action anti-malarial agent being developed in collaboration with the pharmaceutical company Eisai (Nature\, 2017). His most recent discovery revealed a novel cell state responsible for the ability of cancers to resist a wide range of therapies\, and a means to target the cancer therapy-resistant state (Nature\, 2017). His research has been reported in over 650 publications (H index = 147; Schreiber Publications) and recognized through numerous awards\, most recently the Arthur C. Cope Award and the Wolf Prize in Chemistry. \nFour new anti-cancer drugs that target proteins discovered in the Schreiber laboratory have been approved by the U.S. FDA: temsirolimus (Wyeth) and everolimus (Novartis)\, which target mTOR (discovered using rapamycin in 1994)\, for renal cancer\, and vorinostat (Merck) and romidepsin (Celgene)\, which target HDACs (HDAC1 discovered using trapoxin in 1996)\, for cutaneous T-cell lymphoma; in addition\, topical HDAC inhibitor remetinostat (Medivir\, PII clinical trials for CTCL)\, was conceived and synthesized in the Schreiber lab. A small-molecule molecular-glue drug (AP1903) reversed the effects of graft-versus-host disease in acute leukemia patients receiving hematopoietic stem cells engineered to express caspase-9 fused to a drug-responsive\, FKBP12-based dimerization domain (NEJM\, 2011). Proteins first shown by Schreiber to be targeted by a small molecule have been validated therapeutically by the FDA-approval process: tacrolimus (calcineurin/immu­nosuppres­sion/1994; Schreiber’s study of FK506) and bortezomib (proteasome/multiple myeloma/2003; Schreiber’s study of lactacystin). \nSchreiber extended chemical biology principles to medicine by participating in the founding of ten biotech companies\, beginning with Vertex Pharmaceuticals\, whose efforts have made cystic fibrosis a manageable disease. These companies have developed many novel therapeutic agents that are being tested in human clinical trials or used as FDA-approved drugs including: Vertex Pharmaceuticals (founded 1989: fosamprenavir/Lexiva; telaprevir/Incivek; ivacaftor/Kalydeco)\, ARIAD Pharmaceuticals (founded 1991: ponatinib/Iclusig; brigatinib/Alunbrig)\, ARIAD Gene Thera­peutics (founded 1994: ridaforolimus; AP1903)\, and Infinity Pharmaceuticals (founded 2001: reta­spimycin; duvelisib). DESCRIPTION:The 19th Annual Koch Institute Summer Symposium will be held virtually on September 17\, 2021 and will focus on cancer immunotherapy. \nCancer immunotherapy has revolutionized the landscape of cancer treatment\, our thinking of tumor biology and clinical practice. Following the groundbreaking successes of checkpoint blockade therapy and CAR T cell therapy\, culminating in multiple FDA-approved treatments and the awarding of the 2018  Nobel Prize in Medicine to Jim Allison and Tasuku Honjo\, the field is currently at a critical juncture. \nWhile checkpoint blockade therapy has demonstrated that the immune system can be harnessed to fight cancer\,  the next generation of treatments will require us to understand what causes resistance in non-responders\, how this can be overcome\, and how these issues are best addressed clinically. Discussing these questions will be at the core of this symposium as we move towards our ultimate goal to increase the number of patients benefiting from immunotherapy.
